چکیده

Borage (Echium amoenum fisch) is one of the most commonly used medicinal plants, and has long been used as a traditional herbal medicine for many (non)infectious diseases in Iran. Study on photoredox and photoimmunology of borage is little. Natural immunomodulatory plants with minimal adverse/toxic effects could help boost animal health and, ultimately, public health. To determine the effect of borage on the functions of key circulating innate immune cells, effects of borage extract (BE) on bovine neutrophils (PMN) photoredox and phagocytosis events were evaluated using an in vitro model system. Blood PMN isolated from healthy high yielding dairy cows (n = 8/treatment) were pre-incubated with BE and the impact on phagocytosis-dependent-and-independent cellular chemiluminescence (CL), phagocytosis, killing of Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli), fluorescence-based PMN H2O2 production and necrosis were assessed. Relative to control (no BE) PMN, treatment with BE significantly increased phagocytosis-dependent-and-independent PMN CL (>10-15% increase). While BE also led to increased PMN H2O2 production, necrosis was also surprisingly higher in these cells. Phagocytosis and killing of both E. coli and S. aureus by PMN treated with BE was substantially higher than that by control PMN. The increased photoimmunobiological events especially intracellular CL, intracellular H2O2 formation, and phagocytic capacity of BE-treated PMN support the potential immunotherapeutic implications of borage and its components for particularly immunocompromised animals and humans.

Protective Effect of Borage Seed Oil and Gamma Linolenic Acid on DNA: In Vivo and In Vitro Studies

Borage (Borago officinalis L.) seed oil has been used as a treatment for various degenerative diseases. Many useful properties of this oil are attributed to its high gamma linolenic acid content (GLA, 18:3 ω-6). The purpose of this study was to demonstrate the safety and suitability of the use of borage seed oil, along with one of its active components, GLA, with respect to DNA integrity, and t...

Effect of Demecolicne Treatment on The Developmental Competency of In Vitro Matured Bovine Oocytes

Purpose: The aim of this study was to investigate whether demecolicne treatment of matured bovine oocytes adversely affects the process of in vitro fertilization and embryo development. Materials and Methods: Bovine Cumulus Oocyte Complexes (COC's) were matured in vitro and then were randomly allocated to two treatment groups of common concentrations of demecolicne (0.05 and 0.4 mg/ml for 30 mi...

Highly Efficient In Vitro Production of Bovine Blastocyst in Cell-Free Sequential Synthetic Oviductal Fluid vs. TCM199 Vero Cell Co-Culture System

Background The aim of this study was to establish a cell-free sequential culture system that can support high levels of in vitro embryo development and blastocyst formation from bovine zygotes. To this end, this investigation was carried out to evaluate the effects of glucose, serum and EDTA on bovine zygote in vitro development. MaterialsAndMethods Bovine presumptive zygotes were derived from ...

IN VITRO STUDY OF AN ENDOGENOUS IMMUNOSUPPRESSOR FACTOR DERIVED FROM HUMAN OR BOVINE SERUM

The effects of the human and bovine LSF (derived from sera) as well as their purified fractions were studied on murine lymphocytes reactions indicated by blast transformation (BT) assay, mixed lymphocyte culture (MLC) and IgG synthesis. The results indicated that bovine lipid suppressor factor (LSF) has significant immunosuppressive activity on lymphocytes proliferation both in BT and MLC ...

In Vitro Cytotoxic Activity and Binding Properties of Curcumin in the Presence of β-Casein Micelle Nanoparticles

Curcumin (CUR) is the active curcuminoid with many physiological, biochemical, and pharmacological properties. Solubility and stability of CUR is the limiting factors for realizing its therapeutic potential. Bovine β-casein is an abundant milk protein that is highly amphiphilic and self-assembles into stable micellar nanoparticles in aqueous solution. β-Casein nanoparticle can solubilize CUR mo...
